非人灵长类动物(猴子、猩猩,等)在种系上最为接近人类,研究结果更能推演到人类。如果你准备采用这类动物,提醒你要特别注意伦理方面的规定。

一、猴或狒狒动脉粥样硬化模型造模的特点


猴或狒狒作为人类动脉粥样硬化模型动物,具有以下方面的表现:

(1)猴的血脂代谢接近人类,血浆胆固醇主要在LDL中。

(2)血浆脂蛋白对模型饲料的反应与人类相似,模型饲料导致的血脂异常与动脉粥样硬化高度关联。

(3)动脉粥样硬化容易发生,甚至可以自发发生。

(4)模型饲料诱导的动脉粥样硬化,发生于主动脉及其所有大的分支(如,冠状动脉和颅内脑动脉),可以发展到动脉粥样硬化高级阶段,并且容易发生致命性心肌梗塞,这些都是人类发生动脉粥样硬化的类似表现。

二、动脉粥样硬化模型造模中猴或狒狒的选择和使用方法


猴作为人类动脉粥样硬化的模型动物开展研究,需要特别注意猴的选择和使用方法:

1.一般使用旧大陆猴(old world monkey),不使用新大陆猴(New World monkeys)

旧大陆猴包括猴科成员(长尾猴、狒狒等),也可以包括长臂猿科(Hylobatidae)、猩猩科(Pongidae)。

常用于动脉粥样硬化模型的猴包括猕猴(恒菏猴,rhesus monkey)、松鼠猴(squirrel monkey),食蟹猴(cynomolgus monkey,cynomolgus macaques),卷尾猴(Cebus albifrons),也有使用伶猴(titi)。

新大陆猴(例如,卷尾猴,阔鼻猴),由于动脉粥样硬化引起严重的慢性肾脏疾病,后者又影响血脂代谢和动脉粥样硬化,因此,动物容易发生严重病态甚至死亡,因此,这些动物不应当用于动脉粥样硬化模型的造模。猪能够自发发生动脉粥样硬化,对照喂养的猪正常饲料要求较高,需要使用符合要求的普通饲料。

2. 不同种类的猴或狒狒之间,或者相同猴之间,对动脉粥样硬化的敏感程度或者反应性不同,需要进行敏感性(反应性)测试

即便是相同的猴,相同的性别,不同个体对动脉粥样硬化的反应性不同,又由于每组动物数不可能大,因此,应当避免因为个体之间反应性不同而导致的统计学分析结果的不确定性。

为此,可采取的方法是,需要应用模型饲料进行喂养试验,观察血浆胆固醇浓度的改变情况,根据动物的反应性分为高反应性组和低反应性组。或者,只选用高反应性的动物。

3. 动物的年龄和性别

除非研究中有特殊要求,一般使用成年、雄性的猴或狒狒。
