Choline-Deficient Ethionine-Supplemented (CDE)Diet-Induced Oval Cells from mouse Liver

在大鼠使用CDE诱导肝脏干细胞(hepatic progenitor cells)的方法已经比较成熟和稳定之时,小鼠一直没有比较好的方法,因为按照大鼠的剂量转换成小鼠的剂量后死亡率很高,一般不超过2周几乎全军覆没。降低乙硫氨酸浓度虽然能减少死亡甚至不死亡,但诱导的肝脏干细胞非常少。而降低胆碱缺乏程度并且不改变乙硫氨酸浓度,则小鼠不仅状况良好而且在3周左右能够诱导出干细胞44%左右。

【饲料和方法】以下方法之一:

模型饲料代码 方法说明
TP 0100GC50 饮水中含乙硫氨酸0.15%,直接喂养。
TP 1R806CD 与普通饲料混合后喂养,饮水中添加乙硫氨酸0.15%

(1)采用含胆碱50%的胆碱缺乏模型饲料,饮水含乙硫氨酸0.15%

(2)购买无胆碱的胆碱缺乏模型饲料,饮水含乙硫氨酸0.15%

【乙硫氨酸】Sigma公司 No #E5139;Acros公司,等。

【实验动物】小鼠,年龄不限,但最常用成年或接近成年。

【喂养时间】3周或更长时间。
