Choline-Deficient Ethionine-Supplemented (CDE)Diet-Induced Oval Cells from Rat Liver

大鼠是最先采用CDE模型饲料诱导肝脏干细胞( hepatic stem/progenitor cells)的动物。该法采用乙硫氨酸(DL-ethionine)损伤肝细胞,引起的损伤性反应中包括干细胞的反应性增加,从而通过分离、纯化、鉴定而获得肝脏的干细胞。

(1)含0.1%乙硫氨酸的胆碱缺乏饲料(CDE模型饲料)

(2)胆碱缺乏饲料,饮水中加乙硫氨酸

乙硫氨酸一般用0.1%, 0.15%,。

(3)胆碱缺乏饲料,灌胃。

剂量是:按照动物每天饲料饲料量计算,相当于饲料量的0.05~0.1%,或者每天每kg体重50-70mg。(Kaplanski,等)

【乙硫氨酸】Sigma公司 No #E5139;Acros公司,等。

【实验动物】大鼠,年龄不限,但最常用成年或接近成年。

【喂养时间】6周左右。
